The three weeks are flexible, and you can explore Tanzania also in two weeks. Although, if you are interested in different Nationalparks, you might need more time. Those offer 2-3 day tours, and on top of that, you have to travel long distances which also requires some extra time.

Here is our one-month travel route through Tanzania. Of course, you can easily compromise it to 3 weeks, depending on the length of your stay at the Serengeti or other Nationalparks in this beautiful country.
Stop 1: Tanga - three days
Beaches, sun, cocktails, and a lot of street food.
Stop 2: Daressalam - six days
Lively and busy, with many dusty, bumpy roads and lots of Boda Bodas. This city has a general beach area with live music, pubs, and bars for perfect nightlife.
Stop 3: Arusha - two days
It is the start point for the Serengeti safari. Local life, here are no special activities I can recommend. Still, it offers excellent restaurants, street stores, and cheap guesthouses.
Stop 4: SAFARI trip -> Serengeti Nationalpark & Ngorongoro Crater & Tarangire

Stop 5: Dodoma - three days
Like most of the cities in Tanzania, Dodoma is not touristic and is mainly for local life. However, you can visit street markets and eat tons of street food.
Stop 6: Zanzibar - four days
The famous island for relaxing and the "real" travelers. Very touristic but beautiful with beach life, rooftop bars, and an old historic city center.
